Analysis of nuclear DNA content in monosomic maize plants

Five maize monosomics were analyzed for their nuclear DNA content. Several hundred seeds of R/r-X1 X mutants were grown for two weeks and monosomics with missing chromosomes were identified based on their mutant phenotypic characteristics. Nuclei were isolated from both normal and monosomic two-week old seedlings, and stained with 4,6-diamidino-2-phenylindole (DAPI) and propidium iodide (PI) fluorochromes. Nuclei isolated from two-week old seedlings of sorghum were used as an internal standard in the flow analysis. The analysis was carried out on a laser-based Coulter EPICS 750 series flow cytometer-cell sorter. The results indicated lower fluorescence intensities in all the five maize monosomics. In some of the monosomics, the sample size was too small to obtain statistically significant differences in nuclear DNA content between the control and monosomic lines. The results observed in this study suggested that either fluorochrome may be used for detecting monosomics in maize.
